چکیده

We report that erythroid-potentiating activity (EPA). known to stimulate the proliferation of normal human erythroid precursors in vitro. has a growth-promoting effect on human K562 erythroleukemia cells and Friend mouse erythroleukemia cells. Detailed studies were carried out using an EPA produced by a human T-lymphoblast line (Mo). Although EPA has not been purified to homogeneity. several observations indicate that the factor elaborated by Mo cells that stimulates erythroleukemia cell growth is the EPA molecule. The erythroleukemia growth factor cofractionates with EPA using gel exclusion chromatography. isoelectric focusing. and ion exchange chromatography. In addition. the activities exhibit similar

Metalloproteinase Inhibition and Erythroid Potentiation Are Independent Activities of Tissue Inhibitor of Metalloproteinases - l

Tissue inhibitor of metalloproteinases-l (TIMP-l), the major physiological matrix metalloproteinase inhibitor and a potent antimetastatic factor, also stimulates the growth of erythroid progenitors (erythroid-potentiating activity). We analyzed the relationship between the growth factor activity and protease inhibition by preparing purified TIMP-1 “knockout” proteins lacking in vitro antiproteo...

K562 cells produce and respond to human erythroid-potentiating activity.

Human erythroid-potentiating activity (EPA) is a 28,000 mol wt glycoprotein that stimulates the growth of erythroid progenitors in vitro and enhances colony formation by the K562 human erythroleukemia cell line. EPA has potent protease inhibitory activity, and is also referred to as tissue inhibitor of metalloproteinases (TIMP). We observed that colony formation by K562 cells in semi-solid medi...

In vivo effect of human erythroid-potentiating activity on hematopoiesis in mice.

Endotoxin-free purified recombinant human erythroid-potentiating activity (EPA) was administered to normal and bled mice. In anemic ICR mice EPA treatment led to a significant increase in the number of reticulocytes in the peripheral blood and erythroid precursors in the spleen. Stimulation of CFU-E and BFU-E in the spleen was also observed in C3H/HEJ mice, which excluded the possibility of end...

Production of erythroid potentiating factor(s) by a human monocytic cell line.

U-937 is a human monocytic cell line that has been to elaborate factors that affect normal human hematopoiesis in vitro. Studies on the effects of these factors demonstrated an erythroid potentiating factor (EPF) and a potent inhibitor of granulocyte-macrophage (CFU-GM) colony growth. The EPF was present in both serum-containing and serum-free U-937 conditioned media, had a dose-dependent effec...
